چکیده

THE CURRENT METHOD to treat end-stage hepatic disease is whole-liver transplantation. However, this method is limited mainly by a shortage of organ donors. Therefore, developing effective therapies for hepatic cell transplantation represents a major aim of medical research to relieve the suffering of many patients. Most experimental studies on liver cell transplantation are carried out with adult hepatocytes, which have proved to be promising in the ability to repopulate the liver and ameliorate hepatic function. In the last years, many experiments have also been performed with fetal hepatocytes, with encouraging results. In this work we transplanted liver stellate cells, the major producers of extracellular matrix components, to study whether it is possible for them to reach and integrate into the liver of rats. These cells were stably transfected to overexpress green fluorescent protein (GFP) to follow their fate in the liver.

Commitment of bone marrow cells to hepatic stellate cells in mouse.

BACKGROUND/AIMS Recently, several cells found within the liver have been reported to derive from bone marrow (BM). This study sought to examine the commitment of BM cells to hepatic stellate cell (HSC) lineage in mouse liver. METHODS We transplanted BM cells from green fluorescent protein (GFP) transgenic mice into age-matched C57BL/J mice. Hepatic nonparenchymal cells were isolated from the ...

STEM CELLS IN HEMATOLOGY Hematopoietic origin of hepatic stellate cells in the adult liver

Hepatic stellate cells are believed to play a key role in the development of liver fibrosis. Several studies have reported that bone marrow cells can give rise to hepatic stellate cells. We hypothesized that hepatic stellate cells are derived from hematopoietic stem cells. To test this hypothesis, we generated chimeric mice by transplantation of clonal populations of cells derived from single e...

Monocytes Infiltrate the Pancreas via the MCP-1/CCR2 Pathway and Differentiate into Stellate Cells

Recent studies have shown that monocytes possess pluripotent plasticity. We previously reported that monocytes could differentiate into hepatic stellate cells. Although stellate cells are also present in the pancreas, their origin remains unclear. An accumulation of enhanced green fluorescent protein (EGFP)(+)CD45(-) cells was observed in the pancreases and livers of chimeric mice, which were t...

Human Menstrual Blood‐Derived Stem Cells Ameliorate Liver Fibrosis in Mice by Targeting Hepatic Stellate Cells via Paracrine Mediators

Mesenchymal stem cells (MSCs) may have potential applications in regenerative medicine for the treatment of chronic liver diseases (CLDs). Human menstrual blood is a novel source of MSCs, termed menstrual blood-derived stem cells (MenSCs). Compared with bone marrow MSCs, MenSCs exhibit a higher proliferation rate and they can be obtained through a simple, safe, painless procedure without ethica...
